抗體偶聯藥物“ADC”作用機制
ADCs是單克隆抗體和細胞分子的偶聯物,兼具抗體的較高選擇性、較好的穩定性及藥代動力學特征和細胞分子的高*性。
ADCs可分為三部分: 單克隆抗體( monocl onal anti bodi es,mAbs) 、細胞*分子( warhead or payl oad) 和連接基( 1i nker) 。
ADCs對細胞的作用機制
ADCs以靜脈注射的方式進入血液循環, 能避免被胃酸和蛋白水解酶破壞。 通過血液循環進入**組織后, 與**細胞表面的抗原結合,在受體介導下通過網格蛋白( clathrin) 的牽引發生細胞內吞, 形成核內體,氫離子流入核內體中形成酸性環境, 有利于新生的Fc受體與ADCs的mAbs部分結合, 其中部分ADCs與FcRns結合后被排出細胞外, 在堿性環境下, 釋放出ADCs, 重新進入體循環,隨后核內體與溶酶體發生融合, 未被排出細胞外的ADCs在溶酶體的降解下,釋放細胞分子到細胞質中。 細胞分子與DNA、微管蛋白等結合后, 影響其復制或有絲分裂, 導致細胞凋亡。
